Children's Hospital of Philadelphia researchers shed light on genetic disorders tied to cohesin protein malfunction

Researchers studying rare genetic disorders have uncovered insights into those diseases in biological structures that regulate chromosomes when cells divide. Focusing on the cohesin complex, a group of proteins forming a bracelet that encircles chromosome pairs, scientists have discovered mutations that disrupt cohesin, causing a recently recognized class of diseases called cohesinopathies.

"We are learning more about how these genetic abnormalities that affect cohesin play a role in human development," said study leader Matthew A. Deardorff, M.D., Ph.D., a specialist in pediatric genetics at The Children's Hospital of Philadelphia's Center for Cornelia deLange Syndrome and Related Diagnoses.

The research, carried out in children, cell cultures, and zebrafish, appeared May 24 in the American Journal of Human Genetics. Deardorff's co-study leader was Frank J. Kaiser, Ph.D., of the University of Lubeck in Germany.

The cohesin complex is already known to be involved in Cornelia deLange syndrome (CdLS), a multisystem genetic disease affecting an estimated 1 in 10,000 children. The disease has a range of severity, but classically includes mental retardation, impaired growth, heart defects, feeding problems, deformed arms and hands, and distinctive facial features.

Researchers at The Children's Hospital of Philadelphia previously were the first to discover gene mutations that cause CdLS, including forms of the disease with mental retardation and often severe limb abnormalities. The current study identified another gene, RAD21, that when mutated, causes very mild cognitive and physical impairments.

The study team first performed a genome-wide analysis of 101 children with typical CdLS and 189 children having overlapping features of the disease. None of the children had mutations in the three genes already known to cause CdLS. They identified a six-year-old boy with a deletion in a section of chromosome 8 that contains the RAD21 gene, which was known to express a cohesin protein but not previously known to cause disease. As an infant, the boy had been diagnosed with facial features similar to those of CdLS, and subsequently experienced growth retardation, but had normal cognitive development.

The researchers then focused on three additional children with deletions in RAD21 and two children with mutations within the gene, and found a similar patternphysical features, such as short stature and distinctive facial features, overlapping with some of those seen in cohesin disorders, but with only minor cognitive delays. "These findings suggest that children who are very mildly affected may go undiagnosed," said Deardorff.

The research team did further studies in cell cultures and a zebrafish model to investigate molecular mechanisms involved in cohesin disorders. The cohesin complex includes four proteins that join in a bracelet-like structure that surrounds sister chromatids, the identical pairs that result from chromosome duplication prior to cell division. RAD21, the protein expressed by the gene with the same name, forms a clasp that closes the bracelet. A mutated RAD21 gene weakens that clasp, impairing cohesion's normal abilities to repair damage to DNA.

However, Deardorff added, the lab research does not currently explain the full sequence of molecular events, and further studies will investigate knowledge gaps in the process.

As the cost of whole-genome sequencing is rapidly dropping, Deardorff expects researchers to discover additional genes involved in cohesinopathies, offering further clues to how these diseases function in human development. "For now we can expect that patients with the RAD21 mutation will be less severely affected than those with classical CdLS," he said. "As we better understand the mechanisms of these congenital diseases, we'll continue to seek opportunities to devise more effective treatments."

Bigfoot Family Visits Party Store For Goat Supplies

Everyone loves having a birthday party in their honor and the Bigfoot family is no exception. Due to how close knit the Bigfoots are, however, they continue to celebrate the birthdays of all adults involved in full party mode. In fact, in some Bigfoot clans, the adult parties rival the kids parties in fanfare and decorations.

Finding the ideal party supplies can sometimes be a challenge for Bigfoots, though. In spite of the challenges, over the years, the Bigfoots have evolved enough so they are usually able to find what they need in time to throw one of their infamous parties.

?My family and I are a quiet and peace loving folk so it was really upseting when people would run screaming from us when we came to town to get our party supplies,? opines Berta Bigfoot gently. ?Fortunately, after a few more visits to town, people began to let their curiosities get the best of them. Soon, we were able to locate a party store nearby.?

Says Pearl Tucker, ?I admit I was slightly taken aback by the sight of this quartet of giant fur covered beings that had to bend down to comfortably enter my party store.? The owner of the town?s only party store, Party Supplies Palace, continues,?It wasn?t long, however, before I was put at ease by the Bigfoot family and we got down to the business of discussing their party supply needs.?

Finding new and fresh themes for the parties for the members of the Bigfoot family has been proven to be a challenge for both the family itself and Ms Tucker.

?Since the Bigfoot family has a history of throwing big and elaborate parties for all of its members, they had pretty much exhausted their options for this year?s events. Fortunately, after a few meetings with the family, both here at my shop and at their own home, I was able to get a feel for what they like and want,? Ms Tucker shared. The results were a number of special orders that focused on the theme of this year?s parties: the goat.

Pin the tail on the goat, a goat pinata, and goat themed plates, napkins, table coverings and cups are just a few of the items visible to the party guests who have been fortunate enough to snap up an elusive invitation to one of the Bigfoot family?s parties.

Support US Jobs by Repealing Free Trade Agreements

Goal: Bring jobs back to the US by repealing free trade agreements with low-wage countries.

In late 1993, the North American Free Trade Agreement (NAFTA) was signed into law in the US. It eliminated or reduced tariffs (taxes) on a variety of goods between the US, Canada, and Mexico, thereby allowing goods to be moved between these countries at little to no cost. Advocates claimed that it would be good for the US because it would help boost exports and therefore create new jobs to support the increase. While some jobs in the US were created as a result of NAFTA, a recent report estimates that the US trade deficit with Mexico has resulted in a net loss of about 682,900 jobs in the US. Numerous other free trade agreements exist between the US and other countries, such as China, South Korea, Honduras and Panama.

The biggest problems with such agreements arise when there is a wide discrepancy in the pay of workers, working conditions, or relevant environmental laws between member nations. In the case of NAFTA, the US and Canada are relatively similar in these regards and free trade between them may be more beneficial than harmful. However, average wages in Mexico are much lower than in the US and workers experience poorer conditions on the job. These result in significantly lower costs for producing goods in Mexico, and is one reason why manufacturers like Hershey?s Chocolate have moved production facilities just south of the border.

Essentially, free trade agreements have put US workers with decent pay and working conditions in competition with workers in places like Mexico, China, and Vietnam where workers are paid much less and often work under worse conditions. It would be like holding a marathon where one runner is expected to run 20 miles while the other runners only have to run five, and somehow expecting the 20-miler to stand a chance. It is nonsense and it has cost the US jobs and livelihoods. Perhaps the single costliest relationship has been with China, which has resulted in an estimated loss of 2.8 million US jobs.

Free trade agreements with countries that have significantly lower wages, poorer working conditions, or less restrictive environmental regulations than the US are bad deals for US workers. They encourage companies to move jobs overseas and they slow increases in US wages. Urge congressional leaders to repeal these harmful trade agreements to protect US jobs.

NAFTA and similar free trade agreements have created some jobs in the US, but have caused many more to be shipped overseas. The Economic Policy Institute estimates that we have lost nearly 3.5 million jobs to Mexico and China alone.

Many of our most significant agreements are with countries where wages are much lower, working conditions are worse and environmental protections are weaker. Corporations can make larger profits by moving factories to Cancun and Beijing than by keeping them in Detroit. Our free trade agreements make it possible.

All are good photo opportunities ? whether with a "real" camera or a cellphone.?

How do I make sure photos are in focus?

Often, something is in focus, but not the thing you wanted. Many modern point-and-shoot cameras and some cellphones have a face-detection mode that you can select to recognize multiple faces and optimize the focus (as well as exposure and color) for them.

If you don't have this mode, or you want to focus on something else, adjust the focus point to be more precise.?

Newer iPhones make this easy: Simply point on the screen to the part you want to be sharp.?

On other cameras, you can often change the default ? which makes a compromise so that everything is sort of sharp ? and instead chose a smaller focus point or area.?

They still look blurry. What gives?

You may still get a blurry picture if you are in low light because the camera has to keep the shutter open for a relatively long time.?

The trick here is to minimize movement. If you are buying a new point-and-shoot, get one with optical image stabilizations (OIS), which moves either the lens or the image sensor to compensate for jitters.

After having a discussion in a Facebook group this week in which we were asked; If you were starting online this week, what do you hope someone would tell you?

There were quite a few answers and most were running along the same lines; persistence, consistency, patience, etc.

I started thinking that there surely had to be more that others wished they had been told if they knew then, what they know now as an online marketer or business owner.

What is your advice to someone starting out? What is it that you wished you were told? Do you think it would have made a difference in your choices, success or failure?

I?m sure everyone could benefit from a little free advice to someone coming in new or even someone who has worked online for a while.

There should never be an end to learning for any of us.

Texas Tops Finds From Brazil to Bakken as Best Prospect: Energy

Energy companies in search of oil riches rivaling the biggest finds from?Brazil?to?Angola?are flocking to?Texas?shale, where new wells have triggered a 230- fold increase in crude output in three years.

More than 115 years after a gusher 55 miles (88 kilometers) south of?Dallas?ushered in Texas? first oil boom, U.S. producers such as ConocoPhillips and?Marathon Oil Corp. (MRO)?are counting on the Eagle Ford Shale to boost crude output amid a glut-driven slump in natural-gas prices.

Drilling for oil in the brush-covered plains of south Texas is cheaper and less risky than exploration offshore Brazil, the largest oil find in the Western Hemisphere in 30 years, and more profitable than the remote, rougher terrain of the Bakken Shale in?North Dakota?and?Montana.

?The Eagle Ford is the top basin we have in the world today,?David Roberts, chief operating officer at Marathon Oil, told analysts and investors on a conference call last month.

Surging production in shale formations has transformed the U.S. energy landscape, flooding the market with gas and boosting domestic?oil production?by 14 percent from three years ago after dropping by a third in the previous 17 years, according to Energy Department data. After worries of a global oil shortage drove prices to record highs above $140 a barrel in 2008, politicians and industry executives now are discussing the prospect of the U.S. weaning itself from dependence on imports.

Doubling Down

Marathon Oil and ConocoPhillips both plan to double their production in the Eagle Ford this year.EOG Resources Inc. (EOG), based in Houston, calls the Texas shale play its biggest source of growth, and last month boosted its estimated recoverable reserves there by 78 percent.

Oil production?in the Eagle Ford jumped almost sevenfold in 2011 to surpass 30 million barrels, still less than Bakken production in North Dakota that exceeded 128 million barrels. This year daily oil production in the Eagle Ford is forecast to expand by 200,000 barrels, roughly the same amount as the Bakken, according to estimates by Wood Mackenzie Ltd. cited by Hill Vaden, an analyst with the industry consultant.

The?South Texas oil?fields are winning a larger portion of producers? investment because it?s easier and more profitable to drill there compared to many prospects in the U.S. and in the world. Wells are faster and cheaper to develop, and the formation is located closer to refineries on the U.S. Gulf Coast, lowering transportation costs.

Higher Prices

EOG said it costs about $5.5 million per well in the Eagle Ford, compared with more than $8 million per well in the Bakken, because of different well configurations. An offshore?Gulf of Mexico?well can cost $100 million, said?Brian Uhlmer, an analyst at Global Hunter Securities LLC in Houston.

Deep-water wells can take five months or longer to drill, compared to a couple of weeks for a well in the Eagle Ford, said Brian Cain, a spokesman for?Anadarko Petroleum Corp. (APC)

Producers can get a higher price for their Eagle Ford output than they can in the Bakken. Prices for Texas and?Louisiana (USCRLLSS)?crude this week are as much as about $40 a barrel more than production in the?Bakken (USCRLLSS), according to data compiled by Bloomberg.

?The economics there are absolutely stellar,? said Danny Brown, a general manager who helps oversee Anadarko?s Eagle Ford operations. Anadarko has said it is considering selling its exploration properties offshore Brazil.

Less Political Risk

Texas provides a more stable investment environment compared to many international projects, said?Pavel Molchanov, an analyst at Raymond James & Associates in Houston.

?Clearly, there?s less political risk in Texas than in?Libya, let?s say, or Kurdistan,? he said. Marathon Oil last year had output suspended in Libya during unrest in that country.

The Eagle Ford cuts across a 400-mile swath of southern Texas, according to the Railroad Commission, which regulates oil and gas production in the state. Producers have unlocked the resource using advances in horizontal drilling and hydraulic fracturing, which sends jets of water, sand and chemicals underground to break up rock.

Petrohawk Energy Corp., acquired by?BHP Billiton Ltd. (BHP)?last year, first drew attention to the Eagle Ford when it announced a gas find in 2008, a year when futures for the fuel in?New Yorkaveraged more than $8 per million British thermal units.

Surging Production

Expanded use of fracturing, or fracking, across the U.S. caused a surge in gas output that drove prices to a 10-year low this month of $2.204 per million Btu. Meanwhile, crude in New York has climbed 17 percent since the end of 2010 and is trading for more than $106 a barrel.

While drilling has slowed in U.S. shale gas fields such as the Fayetteville in?Arkansas, development has accelerated in South Texas as producers focus on the formation?s oil-rich geology.

ConocoPhillips (COP), the third-largest U.S. oil company, plans to boost production in the Eagle Ford to about 100,000 barrels of oil equivalent a day by the end of 2012 from more than 50,000 barrels a day in late December.

In last year?s fourth quarter, Marathon Oil closed on purchases in the Eagle Ford for about $4.5 billion, according to a regulatory filing. The company said in February that daily output was about 15,000 barrels of oil equivalent in the formation, with plans for 30,000 barrels a day for the full year of 2012.

ConocoPhillips?rose?0.8 percent to $76.51 at the close in New York. Marathon Oil climbed 0.7 percent to $32.46 and EOG?increased?1.3 percent to $111.73.

The Eagle Ford will help lead a surge in state?drilling permits?that?s on pace to reach 25,000 this year, the most since 1985, said Barry Smitherman, the commission?s chairman.

?It?s by far the most sought-after play anywhere ? not only in this country, but anywhere around the world,? said?Fadel Gheit, an analyst at Oppenheimer & Co. in New York.

A Sanford C. Bernstein report last August estimated Eagle Ford production would reach 1.2 million barrels of oil equivalent a day in 2015, with 750,000 of that being liquids.

?A long-time oil field axiom is that big fields tend to get bigger over time, and that?s certainly the case here,? EOG Chief Executive Officer Mark Papa told investors during a Feb. 17 conference call. ?This continues to be the hottest and highest reinvestment rate-of-return play in?North America.?

Recent work in mice suggested that the immune system is involved in removing beta-amyloid, the main Alzheimer's-causing substance in the brain. Researchers have now shown for the first time that this may apply in humans.

Researchers at the Peninsula College of Medicine and Dentistry, University of Exeter with colleagues in the National Institute on Aging in the USA and in Italy screened the expression levels of thousands of genes in blood samples from nearly 700 people. The telltale marker of immune system activity against beta-amyloid, a gene called CCR2, emerged as the top marker associated with memory in people.

The team used a common clinical measure called the Mini Mental State Examination to measure memory and other cognitive functions.

The previous work in mice showed that augmenting the CCR2-activated part of the immune system in the blood stream resulted in improved memory and functioning in mice susceptible to Alzheimer's disease.

Professor David Melzer, who led the work, commented: "This is a very exciting result. It may be that CCR2-associated immunity could be strengthened in humans to slow Alzheimer's disease, but much more work will be needed to ensure that this approach is safe and effective".

Dr Lorna Harries, co-author, commented: "Identification of a key player in the interface between immune function and cognitive ability may help us to gain a better understanding of the disease processes involved in Alzheimer's disease and related disorders."

Alzheimer's disease is the most common form of dementia and affects around 496,000 people in the UK.

(AP) ? The privately bankrolled Dragon capsule arrived at the International Space Station for a historic docking Friday, captured by astronauts wielding a giant robot arm.

It succeeded in making the first commercial delivery into the cosmos.

U.S. astronaut Donald Pettit used the space station's 58-foot robot arm to snare the gleaming white Dragon after a few hours of extra checks and maneuvers. The two vessels came together while sailing above Australia.

"Looks like we've got us a dragon by the tail," Pettit announced from 250 miles up once he locked onto Dragon's docking mechanism.

"You've made a lot of folks happy down here over in Hawthorne and right here in Houston," radioed NASA's Mission Control. "Great job guys."

NASA controllers clapped as their counterparts at SpaceX's control center in Hawthorne, Calif. ? including SpaceX's billionaire maestro, Elon Musk, of PayPal fame ? lifted their arms in triumph and jumped out of their seats to exchange high fives.

This is the first time a private company has attempted to send a vessel to the space station, an achievement previously reserved for a small, elite group of government agencies. And it's the first U.S. craft to visit the station since the final shuttle flight last July.

The astronauts wasted no time getting the Dragon capsule into position for actual docking to the space station. The unmanned capsule is carrying 1,000 pounds of supplies on this unprecedented test flight.

On Thursday, the capsule came within 1? miles of the space station in a practice fly-by. It returned to the neighborhood early Friday so Pettit, along with Dutch astronaut Andre Kuipers could capture it with a robot arm. First, the capsule went through a series of stop-and-go demonstrations to prove it was under good operating control.

NASA ordered extra checks of the Dragon's imaging systems as the capsule drew ever closer to the space station, putting the entire operation slightly behind schedule. At one point, SpaceX controllers ordered a retreat because of a problem with on-board tracking sensors.

Given that the Dragon is a brand new type of vehicle and this is a test flight, the space agency insisted on proceeding cautiously.

A collision at orbital speed ? 17,500 mph ? could prove disastrous for the space station.

SpaceX ? officially known as Space Exploration Technologies Corp. ? is one of several companies vying for the chance to launch Americans from U.S. soil. That ability ended with NASA's final shuttle flight last summer. To get to the space station, NASA astronauts must go through Russia, an expensive and embarrassing situation for the U.S. after a half-century of orbital self-sufficiency.

President Barack Obama is pushing commercial ventures in orbit so NASA can concentrate on grander destinations like asteroids and Mars. Once companies master supply runs, they hope to tackle astronaut ferry runs.

Musk, who founded SpaceX a decade ago and helped create PayPal, said he can have astronauts riding his Dragon capsules to orbit in three or four years. His Falcon 9 rockets lift off from Cape Canaveral, Fla.

The space station has been relying on Russian, Japanese and European cargo ships for supplies ever since the shuttles retired. None of those, however, can bring anything of value back; they're simply loaded with trash and burn up in the atmosphere.

By contrast, the Dragon is designed to safely re-enter the atmosphere, parachuting into the ocean like the Mercury, Gemini and Apollo capsules did back in the 1960s. Assuming all goes well Friday, the space station's six-man crew will release the Dragon next Thursday after filling it with science experiments and equipment.
